GA3 can be used to find the top and bottom relevant parts of a sample. These values can narrow down the scanned Z-range in Jobs by skipping the irrelevant parts of the sample. This is useful for fundamentally speeding up the sample acquisition.
Typical dynamic Z-Stack scanning procedure
Acquire a low resolution Z-Stack.
Threshold - set the size and position of the bounding box.
Acquire the full-resolution Nyquist Z-Stack.
First of all 3D threshold is applied to the channel where the sample is visible.
Bounds are found for each detected object.
Only the minimum and maximum Z are selected using the Modify Columns action.
Select Top action is used for selecting only the smallest and biggest value in their result.
Records are put into one table using the Join Records action.
GA3 recipe is saved using
.Jobs Explorer (Introduction to Jobs Explorer) is opened and the tasks are dragged into the definition to form the following job.
Capturing is set in the Acquisition > Capture Definition task. To obtain the Step value, Z-Stack >
Define Z-Stack is set. Then the first stack is captured by Acquisition >
Capture inside the Z-Stack >
Z-Stack Loop.
Then the GA3 Processing > General Analysis 3 Parameters are defined using the minimum and maximum results from the GA3 recipe.
Z-Stack > Z-Stack Bottom and Top then uses the GA3 recipe From and To values and sets the Step calculated from the Z-Stack >
Define Z-Stack.
Another Z-Stack > Z-Stack Loop is added allowing to capture the narrowed Z-stack by Acquisition >
Capture. Job can be executed (
Run Job) and only the Z stack series falling into the GA3 threshold are captured and saved.
